from the National Institute of Health to develop this technology and
AlphaGene has applied for a patent to cover this significant invention.
AlphaGene refers to this technology
as Single Library Analysis of Gene Expression (SLAGETM).
A SLAGE library contains
tagged cDNAs from the two mRNA sources to be compared. The source-specific tags
track the identity of each mRNA source throughout the analysis.
The SLAGE
library provides a renewable supply of source-specific probes for repeated
differential gene expression studies from very small amounts of tissue. The
SLAGE method is based on a mixed cDNA pool as the starting material for library
construction. Because, after mixing, the library construction protocol is
identical for both mRNA sources, a true ratiometric comparison can be made and
false positive leads can be minimized.
